Obstructive jaundice due to the reflux of bile into the blood stream is observed under a variety of pathological conditions, with a high incidence of ineffective complications. The basis for this predisposition is not understood, but some data point to the presence of immunosuppression linked with hyperbilirubinemia. in an attempt to elucidate the nascent events in this study we followed up the changes in immune status of mice subjected to common bile duct ligation (CBDL) and investigated the potential therapeutic effects of PGM-Zn, showing immunostimulatory and hepatotropic effects in our previous experiments. The analysis included estimation of phenotype and cell cycle analysis of cells in spleen. Treatment with PGM-Zn (10 mg/kg i. p.)or saline solution started 6 days before CBDL. The data have shown that cholestasis in mice decreases the proportion of splenic CD4, CD5 and Ig+ cells and the proportion of spleen cells in S and G2/M cycle phase. The pre-treatment with PGM-Zn, augmented the initial value of Ig+ cells in the spleen and almost completely abrogated the initial signs of immunosuppression and hypoplasia after CBDL. The data point to CBDL-induced immunosuppression and the therapeutic potential of PGM-Zn.
