Immunological and serological changes in cholestatic jaundice and immunocorrecitve potential of peptidoglycan monomer linked with zinc (PGM-Zn)

Patients with jaundice due to extrahepatic biliary obstruction still experience a high rate of postoperative complications and death. High levels of bile salts and intracellular bilirubin are suspected as mediators of the systemic consequences of obstructive jaundice, but recent data suggest that disturbances in macrophage-immune system interactions may be responsible. This study focused on the immunological, phagocytic and biochemical alterations of BALB/c mice subjected to common bile duct ligation (CBDL) and potential therapeutic effects of PGM-Zn, which in our previous experiments has shown immunostimulatory and hepatotropic effects. Immunological disturbances observed in animals were proved either in patients suffered of cholestatic jaundice. The data have shown that on the first p.o. day in thymus increases the number of single, but decreases the proportion of double positive cells. Subsequently, the number of double negative cells increased. In the spleen during the cholestasis a decrease of proportion of CD4+, CD5+ and Ig+ cells were also seen. The effects were followed by increased number of cells in S and G2/M phases. Pre-treatment with PGM-Zn prolonged the survival of mice with CBDL. Concomitantly, expression of class II in the liver increased, and augmented the proportion of double negative and CD4+CD8- cells in thymus, and CD4+ and Ig+ cells in spleen. Hyperplastic effect of PGM-Zn on thymic and splenic cells was proved by cell cycle analysis. PGM-Zn had also some influences on biochemical alterations characteristic for obstructive jaundice. The data point to CBDL-induced immunosuppression and to therapeutic potential PGM-Zn in these events.
