Expression Pattern of Genes Involved in Hemopoietic Cell Differentiation
Leukemic cells arise from cells stopped at various developmental stages during differentiation. In this context we study the mechanisms of leukemia development by analysing genes involved in hemopoietic cell differentiation.Gene targeting studies in mice show that major players in the commitment of hemopoietic progenitors are transcription factors from the Ikaros and Notch families.Studies of cell populations from patients with leukemia indicate that inappropriate expression of these genes might be involved in leukemogenesis.Accordingly, we analyzed the differential expression of transcription factors and their splicing variants from the Ikaros family, Ikaros, Aiolos and Helios in the most frequent leukemia types, CLL, ALL and AML as well as in hematological cell lines.We used RT-PCR on RNA isolated from hemopoietic cells of healthy individuals and patients previously evaluated by immunophenotypic criteria.Our results show that mRNA expression of Ikaros, Aiolos and Helios varies in our samples, but is not restricted to just one type of leukemia.Further, we provide evidence that splicing variants of these transcription factors occur but they are not restricted to the leukemic cells.
