Protein carbonyl groups in gills of mussel Mytilus galloprovincialis as biomarkers of oxidative stress in marine environment
Oxidative stress, an imbalance toward the pro-oxidant side of the pro-oxidant/antioxidant homeostasis, occurs in the marine organisms as a consequence of environmental factors’ fluctuation and anthropogenic activities/pollution. In the present work we measured changes in different biomarkers of effect and exposure after treatment of Mediterranean mussel Mytilus galloprovincialis with model agents Tributyltin, Hydrogen Peroxide and Water soluble fraction of Diesel oil. For estimation of the risks posed by pollutant discharges to the environment and natural resources, we developed methods for the detection of carbonylated (damaged) proteins and evaluated its applicability for oxidative stress determination. These findings were compared with estimates of other biomarkers (Hsp70 and DNA integrity) used for marine pollution biomonitoring in the frame of the environmental programme “Project Adriatic”. The usage of protein carbonyl groups as biomarkers of oxidative stress has some advantages in comparison with the measurement of other oxidation products because of the relative early formation and the relative stability of carbonylated proteins. The measurement of protein carbonyl groups after their derivatisation with 2, 4-dinitrophenyl-hydrazine (DNPH) is the most widely utilized measure of protein oxidation. Stable dinitrophenyl (DNP) hydrazone product was detected in mussel gills by dot blot immunoassay, quick and suitable method for large numbers of samples analysis. The results indicate causal relationship among high levels of newly formed protein carbonyl groups and two stress biomarkers in mussel treated with model agents and mussels collected at 19 sites along Croatian coast of Adriatic Sea. As expected, induction of Hsp70 proteins followed protein damage measured as formation of carbonyl groups. Concomitantly, both “protein” biomarkers correlate with DNA damage/integrity alterations as a consequence of oxidative stress due to pollutant exposure.
