Variations and interrelation in quantification of mumps virus by two methods based on cytopathic effect
Conventional assays in the virological laboratories for identification of a virus isolate and quantification of infectious virus particles are dependent on the observation of cytopathic effect (CPE). Like in many virus samples the quantification of infectious mumps virus particles is routinely performed by plaque forming unit (PFU) assay and/or 50% cell culture infective dose (CCID50) assay. In this study we analyze variations and interrelation in quantification of nine mumps virus strains by these two methods using standard protocols. Strong intrinsic characteristic of each mumps virus strain to interact distinctively with Vero cell culture results in a unique CPE. Thus the titer values of analyzed mumps virus strains arised from the two methods differed significantly for some strains while were almost identical for others. The results reported here indicate that care should be taken when comparing the numbers of infectious virus particles in virus samples obtained by these methods of quantification.
