Peptidoglycan monomer augments lymphocyte proliferation and IFN- and IL-4 production in ovalbumin immunized mice

Peptidoglycan monomer (PGM) augments several fold humoral immune response to ovalbumin (OVA) in mice after s.c. administration of PGM mixed with OVA. It significantly stimulates IgG1 and IgG2a subclasses of specific antibodies indicating upregulation of both Th1 like and Th2 like immune response1. In this study we describe the influence of PGM on lymph node enlargment, IFN- and IL-4 secretion and proliferative response of OVA-specific lymphocytes. Mice were immunized s.c. with OVA or with OVA+PGM in saline. The cell suspensions of the draining lymph nodes were collected at different time intervals after the booster and cultivated in the presence of OVA. Culture supernatants were screened for IFN- and IL-4 by capture ELISA. Proliferative response were evaluated by pulsing the cultures with 3H-thymidine and measuring the incorporated radioactivity 24 hours later. Groups treated with PGM had always the highest lymph node cellularity. The production of IL-4 and IFN- was upregulated in lymph nodes of PGM treated mice in comparison to those which received only the antigen. Kinetics of IFN- and IL-4 secretion differed, IFN- being the first one to show up and IL-4 arising later. The noticed cytokine secretion was OVA-specific. Cultures of lymph nodes from PGM treated mice showed stimulated proliferation of OVA-specific lymphocytes, suggesting that in those lymph nodes the higher percentage of OVA-specific cells was present in comparison to lymph nodes from mice immunized with OVA alone.
