Brain free fatty acid accumulation in hypoxia and epilepsy
Numerous pathological conditions, including hypoxia and epilepsy, are accompanied by an increase in intracellular phospholipases A2 and C, degradation of membrane phospholipids and liberation of cerebral firee fatty acids (FFA). The aim of this study was to determine accumulation dynamics of the brain FFA in rats exposed to controlled hypoxia and experimental epilepsy. The animals of the first group were subjected to hypoxia procedure and decapitated immediately, 5, 15 and 60 minutes after loosing the righting reflex. The second group received penicillin (5000 U/5microL) into the left lateral ventricle and were decapitated immediately, 5 or 15 minutes after the appearance of epileptic seizures. The brains were quickly removed and lipids were extracted. FFA were separated using TLC and after methylation quantified by gas chromatography. An increase in total brain FFA content and fiee arachidonic acid was detected in hypoxia-exposed rats with the maximum level at 60 min and 15 min respectively after loosing the righting reflex. Seizures produced an increase in all determined FFA reaching a statistically significant level 5 min after their appearance.
