Formation of bone-like apatite layer on hydroxyapatite/gelatin scaffold
A composite of hydroxyapatite (HAp) and gelatin (G) was prepared by the biomimetic approach. In the first stage, complete conversion of aragonitic cuttlefish bone to highly porous hydroxyapatite was achieved. In the second stage, different concentration of gelatin solution (5, 10, 20 wt. %) was impregnated into the HAp scaffolds and cross-linked with glutaraldehyde. In vitro testing was performed in Hank’s balanced salt solution (HBSS) for period up to 28 days at 37°C. The surface of composite samples was investigated using scanning electron microscopy (SEM) attached with energy dispersive X-ray spectroscopy (EDX). The SEM-EDX analysis of the composite surface showed the precipitation of calcium deficient hydroxyapatite (CDHA) and amorphous calcium phosphate (ACP) depending on the nature of precipitation substrate. The chemical analysis of HBSS solution, performed after in vitro testings by ionic chromatography method, also indicated calcium phosphate precipitation on the composite surface. These materials are of great interest for bone regeneration applications due their ability to form bonelike apatite when implanted in vivo.
