Lamotrigine: Influence on the activity of Na+, K+-ATPase, JNK and c-Jun induction in rat hippocampus after kainic acid treatment
We examined the effects of lamotrigine (LTG) on the activity of Na+, K+-ATPase, and stress- activated protein kinase, c-Jun N-terminal kinase (JNK) and induction of c-Jun protein in rat hippocampus after kainic acid (KA) treatment. Enzyme activity from hippocampal tissue was detarmined two hours, three and five days after single subcutaneous KA (8 mg/kg) injection. The activity of Na+, K+-ATPase is also determined in rats pretreated with LTG (30 mg/kg) two hours before KA application. Hippocampal c-Jun and JNK induction were determined two hours and five days after KA treatment and also in LTG- pretreated rats. Na+, K+-ATPase activity was significantly inhibited the third and fifth day after KA application. C-Jun and JNK induction were observed two hours and five days after KA treatment. In the group of LTG- pretreated rats Na+, K+-ATPase inhibition, c-Jun and JNK induction were detected the fifth day after KA treatment. The inhibition of Na+, K+-ATPase observed the fifth day after KA administration was significantly less pronounced in the LTG- pretreated rats. It may be concluded that LTG has a partialy protective effect in KA-treated rats.
