Cytogenetic studies of long-term in vitro cultures of Picea omorika (Panč.) Purk. and Beta vulgaris L.
Cytogenetic studies were carried out on 3-year old embryogenic cell line of Picea omorika (2n=24). Comparative analyses of proembryonal and somatic embryo cells showed that mitotic and chromosomal abberations, e.g. sticky chromosomes, anaphase/telophase bridges, chromosome fragmentations and polyploidy appeared very rarely. Chromosome morphology and heterochromatin distribution after fluorochrome banding were the same as in root-tip cells of seedlings (22 CMA-positive bands out of which 16 bands represent NORs). According to the number of NORs, the maximum number of 16 nucleoli was counted. There was no evidence of morphological changes of proembryos or mature somatic embryos as well as the loss of embryogenic capacity. Karyological analyses were conducted in Beta vulgaris (2n=18) 20-year old habituated organogenic (HO) and nonorganogenic (HNO) callus cultures and callus (NK) obtained on the PG medium with growth regulators, as well as in 6-year old tumours tissue (TT), induced by Agrobacterium tumefaciens. In all tissues studied the cells with different degree of polyploidy were found. The highest level of mixoploidy was observed in HNO callus, while in the HO callus, most of the cells were diploid. The number of nucleoli increased proportionally with the degree of polyploidy. Lagging chromosomes observed indicate the appearance of aneuploidy.
