The influence of nimodipine and nicardipine on the brain free fatty acid level in rats with electroconvulsive shock-induced seizures

Enhanced neuronal activity during epileptic discharge increases the concentration of the free fatty acids (FFAs) in the brain. This biochemical disorder is caused by the increase of the intracellular Ca2+ concentration, the activation of membrane phospholipases C and A2, and by degradation of membrane phospholipids. FFAs have detrimental efiects on cell membranes. Accumulation of their metabolites causes further intracellular brain damage. The aim of this study was to investigate the effect of the calcium channel blockers, nimodipine and nicardipine, on the brain FFA level in rats with electroconvulsive shock (ECS)- induced seizures. The study was carried out on Hannover-Wistar rats. Prior to ECS treatment some rats did not receive any substance, while the animals of control group received vehicle solution. The other rats received tested doses (1, 3 or 10 mg/kg) of mentioned calcium channel blockers. All rats were decapitated 3 min after ECS treatment. The brains were taken out and frozen in liquid nitrogen. The FFAs were quantified using gas chromatography method. The results indicate that ECS-induced seizures cause the increase of the brain FFA level. Tested doses of nimodipine did not influence the increase of the brain FAA level while nicardipine prevented the increase of the free palmitic and arachidonic acid level in rats with ECS-induced seizures. It may be assumed that the brain FFA accumulation caused by ECS treatment is not associated predominantly with a disturbance in Ca2+ homeostasis via L-type voltage-sensitive Ca2+ channels, but by some other mechanisms.
