Calcium channel blockers and the brain free fatty acid level in rats with status epilepticus induced by lithium and pilocarpine administration
Preserved morphological and functional cell membrane integrity is fundamental for normal functioning of the brain cells. Seizure activity, when continued for a sufficient time, induces neuronal death. Seizures trigger a variety of biochemical processes including an inftux .of extracellular Ca2+ through voltage-dependent and glutamate-operated channels, activation of membrane phospholipases, liberation of the free fatty acids (FFAs), diacylglicerols, eicosanoids, lipid peroxides and free radicals. These lipid metabolites, along with abnormal ion homeostasis, may be involved in cell injury and death. The aim of this study was to determine the inftuence of the calcium channel blockers on the brain FFA level in rats with generalised convulsive status epilepticus induced by lithium and pilocarpine administration. Various doses (1, 3 or 10 mg/kg) of nimodipine and nicardipine had been injected i.p. 30 min after administration of pilocarpine (30 mg/kg) to lithium-treated (3 mmol/kg) rats. All rats were decapitated 30 min after the administration of vehicle solution, nimodipine or nicardipine. The FFAs were quantified using gas chromatography method. The results demonstrate that lithium plus pilocarpine- induced status epilepticus causes the brain FFA accumulation. Tested doses of nicardipine did not inftuence the increase of the brain FAA level while nimodipine prevented the accumulation of the brain free palmitic, oleic and docosahexaenoic acid in rats with status epilepticus.
