Genetic determinants of cardiocerebrovascular disease
Cardiocerebrovascular insufficiency involves multifactorial disorders influenced by risk factors such as disturbance of the lipid profile, hypertension and genetic polymorphism. Several genetic loci are involved in the development of the cardiocerebrovascular disease. In this study, we investigated apo E and ACE polymorphisms in 60 patients with myocardial infarction and 60 patients with angiographically verified cerebral artery disease. The results revealed 20% of the patients to be E4/4 homozygotes, indicating an increased prevalence of E4/4 homozygosity as compared to control subjects (2%), and suggesting its probable atherogenic role. The study also demonstrated a positive correlation between apo E4/4 and the level of plasma Lp(a), cholesterol, LDL cholesterol and DD allele. The frequencies of the I and D alleles in patients were 0.28 and 0.72, and in control population 0.47 and 0.53, respectively. The findings suggest that MI and cerebral atherosclerosis are partially determined by genetic disposition. Increased proportion of apo E4/4 and ACE D/D genotypes observed in patients might be of prognostic significance in the development of cardiocerebrovascular complications.
