Oksidacija fenolnih spojeva katalizirana vlastito proizvedenom lakazom iz Trametes versicolor
Gljiva bijelog truljenja Trametes versicolor proizvodi tri lignolitička enzima: lakazu, lignin peroksidazu i mangan peroksidazu te posjeduje sposobnost razgradnje lignina, policikličkih aromatskih ugljikovodika i ostalih organskih sintetskih spojeva. Lakaze (EC 1.10.3.2) su oksidaze koje kataliziraju oksidaciju supstrata prijenosom elektrona i istovremenom redukcijom kisika u vodu. Cilj ovog rada bio je proizvodnja lakaze iz Trametes versicolor G-99 u submerznim uvjetima na otpadu iz industrije papira, te oksidacija fenolnih spojeva: fenola, katehola i koniferilnog alkohola, katalizirana vlastito proizvedenom lakazom. Fenolni spojevi su široko rasprostranjeni spojevi u prirodi. Sastavni su dio industrijskih otpadnih voda kao npr. fenol i katehol. S druge strane, mogu biti dio stanične stijenke biljaka kao koniferilni alkohol. Pokusima enzimske oksidacije u kotlastom reaktoru ostvarene su 100%-tne konverzije fenola, katehola i koniferilnog alkohola. Razvijeni matematički model procesa dobro opisuje eksperimentalne podatke.

Oxidation of phenolic compounds catalyzed by own produced laccase from Trametes versicolor
Trametes versicolor is a white-rot fungus that produces three ligninolytic enzymes: laccase, lignin-peroxidase and manganese-peroxidase, and has an efficient degradation capacity of lignin, polycyclic aromatic hydrocarbons and a number of synthetic dyes. Laccases (EC 1.10.3.2) are multi-copper oxidases that catalyze oxidation of substrates by electron transfer and simultaneous reduction of oxygen to water. In this work, Trametes versicolor MZKI G-99 was grown in a submerged culture using waste from paper industry with the aim of laccase production. Once produced, laccase was kinetically characterized and used as a catalyst in the oxidation of three different phenolic compounds: phenol, catechol and coniferyl alcohol, without any purification and concentration steps. Phenolic compounds can be found in numerous industrial wastewaters (phenol and catechol). They can also be the main constituents of the plant cell walls (coniferyl alcohol). The experiments were carried out in a batch reactor and 100% conversion of laccase-catalyzed oxidation of phenol, catechol and coniferyl alcohol was obtained. Mathematical model of the process was developed and fits the experimental data well.
