Alteration in mice adipose tissue phospholipids composition induced by dietary fatty acid variation

Phospholipids (PLs) composition of various body tissues is susceptible to modifications by varying the composition of dietary fats. The aim of this study was to determine the effects of five different diets, as a source of different groups of fatty acids (FAs), on mice adipose tissue (AT) PLs composition. Beside the standard diet, the pumpkin-seed oil (as source of n-6 and n-9 FAs), olive oil (n-9 FAs), lard (n-9 and saturated FAs) and fish oil enriched diet (n-3 FAs) were used for feeding wild type C57Bl/6 male mice aged 8-10 weeks for 3 weeks. The composition of different class of PLs and the FA chains of total PLs were analysed in mice groups fed with different diet types. AT total lipids were extracted by the method of Folch. PLs were quantified spectrophotometrically after the separation by two-dimensional TLC. FAs were analysed by GC. Analysis of the PL classes showed that phosphatidylcholine (PC), phosphatidylethanolamine (PE), phosphatidylinositol and phosphatidylserine were common components in all diet types. In standard diet group as in fish oil enriched diet group phosphatidic acid was determined, while in other enriched diet groups lysophospholipids were determined. The main constituents of all analysed diet types were PC and PE. The resulting ratio of these two components had the lowest value in lard (1.8), and the highest in the pumpkin seed oil enriched diet group (3.6). FA compositions of PLs in all diet types were characterised by a preponderance of C18 acids and among them stearic acid was the main FA. Additionally, in all diet types unsaturated FAs prevailed with oleic and arachidonic acid as the main FAs. Generally, FA composition of tested PLs slightly follows the composition of applied diets’ FAs. AT metabolism is well known and explored but less is known about the function and composition of AT PLs. The data obtained in this research could help realize the role of PLs in dietary altered AT metabolism.
