Co-cultivation of Legionella and amoeba Dictyostelium discoideum
Legionella naturally exploits numerous species of protozoa, which provide nutrients, protect the bacteria from adverse conditions and serve as a vehicle for the colonization of new habitats. D. discoideum is a unicellular, free-living amoeba that lives in soil and water and feeds on bacteria. This report describes the conditions of co-cultivation of Legionella and amoeba D. discoideum. Infection of D. discoideum with Legionella requires an incubation temperature of 25°C. Amoeba cells grown axenically were cultured in HL-5 liquid medium. Legionella were harvested after 48 h growth on BCYE plates, resuspended in water or PBS, and used to infect D. discoideum at an MOI of 10:1. Bacteria and amoeba was co-cultivated in two different media: HL5 media and assay media. The number of viable bacteria on each day was determined by measuring CFU. One day after co-cultivation significantly lower number of L. longbeachae was detected (~102) in comparisom to number of L. pneumophila (~104). The number of L. pneumophila increased during 96 hours when ~105 CFU/well was detected in bouth media. L. longbeachae was able to grew better in assay media and 96 h p.i. ~104 CFU/well was detected. In HL5 media the number of L. longbeachae increased only for 0, 5 log. Importantly, HL5 medium alone or assay media does not support L. pneumophila or L. longbeachae growth, indicating that all growth detected is a result of intracellular multiplication. In conclusion, bouth Legionella are able to infect and proliferate within D. discoideum and replication was better in assay media.
