The effect of peptidoglycan monomer on cytokine of T-cell responses to ovalbumin in mice

Peptidoglycan monomer (PGM) originating from Brevibacterium divaricatum posses immunomodulating properties. We have shown recently that humoral immune response to ovalbumin (OVA) in mice could be potentiated several fold after s.c. administration of PGM mixed with OVA. The analysis of IgG isotype compositin showed that IgG1 and IgG2a were significantly stimulated, indicating upregulation of both Th1 and Th2 like immune response. In this study the T cell activation was monitored as secretion of IL-4 and IFN-gamma from lymph nodes lymphocytes of immunized CBA mice. Mice were immunized s.c. with OVA and with OVA+PGM in saline. The cell suspensions of the draining lymph nodes were collected in different time intervals and cultivated in the presence of OVA. Culture supernatants were screened for IFN-gamma and IL-4 by capture ELISA established in the laboratory. Enlargment of lymph nodes of mice treated with OVA and with OVA+PGM was noticed in comparison to control group receiving only saline at all time points tested, but groups treated with PGM had always the highest lymph node cellularity. Measurable amounts of both cytokines were detected in draining lymph nodes fourth day after immunization, the quantities being higher in the groups treated with PGM. The more pronounced effect of PGM on cytokine secretion was noticed 12 and 36 hours after the booster. Again the production of IL-4 and IFN-gamma was upregulated in immunized mice treated with PGM in comparison to mice which received only the antigen. Kinetics of IFN-gamma and IL-4 secretion differed, IFN-gamma being the first one to show up (12 hrs after the booster), and IL-4 arising later (36 hrs after the booster). The noticed cytokine secretion was OVA-specific since no IFN-gamma or IL-4 was found in the lymph nodes cultures of control mice treated with saline. These results corroborate our earlier findings that PGM stimulates both Th1 and Th2 like immune responses.
