A Continuous Spectrophotometric Assay for Measuring Amino Acid Activation by Peptide Synthetases
Background and purpose: Peptide synthetases are multifunctional enzyme complexes catalysing the non-ribosomal biosynthesis of a variety of bioactive peptide products. The activation reaction is generally monitored using the isotope exchange assay, which suffers several disadvantages such as discontinuous monitoring and the use of radioactive reagents. A continuous spectrophotometric assay was introduced for the measurement of PPi generation accompanying the amino acid activation reaction catalysed by peptide synthetases.
