BMP signaling pathway in experimental inflammatory bowel disease
Bone morphogenetic protein-7 (BMP-7) is a member of BMP family which belongs to the transforming growth factor-beta (TGF-beta) superfamily. BMP-7 induces bone formation and regeneration and determines important steps during bone development. In addition, BMP-7 is used as therapeutic agent for bone repair especially for fracture healing. Beside its osteoinductive properties, BMP-7 may also involved in development and regeneration of non-skeletal tissues and organs such as kidney, heart, intestine and brain. BMPs efforts their effect by activation of specific receptors (BMPR) leads to activation of the BMP signaling pathway, namely intracellular Smad proteins. Alterations in BMPs expression and BMP signaling pathway have been associated with the pathological conditions linked to several human diseases such as inflammatory bowel disease (IBD) and colon cancers. We recently described the expression of different BMPs including BMP-2, -4, -6 and -7 in colon tissue during experimental IBD. The aim of this study was to investigate the BMP receptors and BMP signaling pathway including R-Smads (Smad1, Smad5 and Smad 8), Co-Smad (Smad4) and I-Smads (Smad6 and Smad7) in IBD by the continuous monitoring of the expression these signaling molecules in rat colons during different phases of experimental IBD as well as after BMP-7 treatment. IBD was induced by intrarectal administration of trinitrobenzenesulfonic acid (TNBS). After the IBD induction, the rats were treated with BMP-7 (100 g/ml) and sacrificed on the 2nd, 5th, 14th, and 30th day after the TNBS induction. Expression levels Smad proteins were determined by RT-PCR analyses. Our results show that BMP pathway is active in IBD as well as after BMP-7 treatment. The expression of all three BMP receptors including BMPR type I (IA and IB) and BMPR type II was found with evident constant expression of BMPRIA during disease. The expression of BMPRIB was slightly decreased during disease with significant elevation after BMP-7 treatment while BMPRII expression was higher in chronic stage of disease and became constant after BMP-7 treatment. Further, the increased expression of BMP-specific R-Smads (Smad1, Smad5 and Smad8) was observed during IBD as well as after BMP-7 treatment. Expression of Smad4 as Co-Smad was unchanged during diseases. Furthermore, the expression levels of inhibitory Smads (Smad6 and Smad7) were increased in colon samples from diseased animals in comparison with the colon of animals treated with BMP-7. The presence of BMPs and BMP signaling pathway during IBD suggests an important role of BMPs in the regulation of colon tissue damage and healing during disease. BMPs, in particular BMP-7, may be used to protect intestine during IBD as well as bone which is common complication of long-lasting IBD.
