A Green Route Towards Sustainable Organic Synthesis - Exploiting the Power of Mechanochemistry
Since the beginning of the modern organic synthesis era, one of the great challenges chemists have been faced with is the reduction of environmental hazards associated with the beauty of designing and constructing complex organic molecules. Another equally important consideration in the conventional solution-based synthesis deals with diminished product yields or reactant conversions and the related economic issues due to loss of the starting material. In the search for a preparative method that would meet the requirements of green chemistry and successfully address the abovementioned problems, mechanical treatment of (in)organic reagents by simply grinding them together has reappeared as one of the potential solutions. The emerging field of organic mechanosynthesis provides a clean and environmentally-friendly route to C-C bond formation as well as to many fullerene-based and heterocyclic systems. Particularly interesting reagents are isothiocyanates and isocyanates which have been shown to undergo a quantitative conversion to the corresponding products by manual grinding. In this presentation, recent advancements in the mechanosynthesis of thiourea and guanidine targets achieved by our group will be described. Solvent-free grinding methodology based on either manual treatment or automated ball-milling of the starting material has been applied for the quantitative synthesis of structurally diverse thiourea derivatives. Furthermore, by utilising the concept of liquid-assisted grinding (LAG) we demonstrate its potential for the synthesis of a series of guanidine- and biguanide-derived compounds.
