High resolution structure of the Mn derivative of insulin
This paper presents results of the structural investigation on the human insulin hexamer derivative stabilised by manganese coordination. Single crystals of the Mn-insulin derivative were prepared by the hanging drop vapour diffusion crystallisation method. Due to synchrotron radiation the data were collected to high resolution (1.05 Å). In the structure of the Mn insulin derivative both manganese ions lie on the three-fold axis similarly as in the Zn- and Ni-insulin derivative. Each Mn ion is octahedrally coordinated by three symmetrically related His and three water molecules. Positions of atoms are better defined by the electron density in the Mn derivative in comparison with the structure of the Ni derivative at 1.35 Å resolution.
