Binding of cationic polyamines with double-stranded polynucleotides studied by surface-enhanced Raman spectroscopy

Surface-enhanced Raman scattering (SERS) was used to study interactions of DNA binding compounds, PHENPOD and PYPOD, with double-stranded polynucleotides, poly dAdT-poly dAdT and poly dGdC-poly dGdC. SERS spectra of solutions at low polyamine concentrations (10–6–10–5 mol dm3) revealed strong enhancement of the Raman scattering from the polyamine molecules attached on the colloidal silver particles. New bands observed in the spectra with double-stranded polynucleotides originated mainly from nucleobases, indicating interactions between polyamines and polynucleotides. It was obtained that PHENPOD and PYPOD selectively interact with ace-enhanced Raman scattering (SERS) was used to study interactions of DNA binding compounds, PHENPOD and PYPOD, with double-stranded polynucleotides, poly dAdT-poly dAdT and poly dGdC-poly dGdC. Interactions of both polyamines with the major and the minor groove are recognized for poly dAdT-poly dAdT, while only the PYPOD molecules bind to the major groove of the poly dGdC-poly dGdC polynucleotide.
