Comparative analysis of the micronuclei incidence of persons occupationally exposed to ionising radiation in Serbia and Croatia
Biomonitoring is important part of biodosimetry for individuals professionally exposed to different physical or chemical mutagens or carcinogens. To estimate the amount of damage caused after the exposure, it is important to have sensitive and reliable methods for evaluation. The reliable method for detection of stabile DNA damage in human peripheral blood lymphocytes in the field of radiation protection is cytokinesis-blocked micronucleus assay (CBMN). Total number of the individuals included in this study was 60, half of them were workers from Serbia and the other half workers from Croatia. The samples of whole blood (5ml) were taken from cubital vene into heparinised vacutainer (Becton Dickinson) and kept at 4 °C till further processing. One thousand binucleated lymphocytes with well-preserved cytoplasm per subject were analyzed. The criteria used for the selection of binucleated cells, MN, NPB (total number of nucleoplasmic bridges), NB (total number of nuclear buds), and for the calculation of NDI (nuclear division index). When two groups were compared, statistically significant difference was observed for MN (p=0.009), MN1 (p=0.046), NB (p=0.005) and NMB (p=0.000). All the values were higher in the group of workers from Serbia. Spearman correlation did not show any statistically significant influence on observed variables, except for the MN and MN1 (R=0.885). The number of NMB also showed the significant correlation with the NB (R=0.623). The interesting fact was the significant correlation between NB and total years of working (R=0.439), but not with the years of exposure, while NMB showed significant correlation with total years of employment (R=0.413) and years of exposure (R=0.264).
