Olive Oil Polyphenols Induced Changes in Glutathione Level Enhance Liver Regenerative Capacity in Mice

Polyphenols may increase the capacity of endogenous antioxidant defences and modulate the cellular redox state which may have wide-ranging consequences for cellular proliferation, as in process of liver regeneration. Because gluthatione is the most important determinant of cellular redox state, modulation of its level may alter cellular processes via the activation of trancriptional factors, such as Nrf2, which regulates expression of gamma-glutamate-cysteine ligase (GSCc), a key enzyme in glutathione biosynthesis. Therefore, attempts were made to understand the regulation of glutathione synthesis in response to olive oil polyphenols extract (PFE) treatment during liver regeneration induced by 1/3 partial hepatectomy (pHx) in mice. Total liver glutathione concentration, RT-PCR mRNA liver levels for GCSc and Nrf2 and the serum level of interleukine-6 (IL-6) which supports the liver regeneration was evaluated in the group of mice treated with physiological solution (FO) or PFE followed by 1/3 pHx, and in the group of mice receiving vehicle alone. Additionally, liver mass restoration was calculated. PFE provoked an initial increase in glutathione level followed by a sharp decrease at 3h after pHx and continuous raise of its concentration in subsequent periods. These changes, implying to the oxidative stress induction, were accompanied by Nrf2 induction in all time intervals, and a higher and prolonged expression level of the GCSc gene in particular in periods accompanied by the glutathione level increment. Although PFE hampered the rise of IL-6 in the serum, this effect didn't impair the liver regenerative ability, as the regeneration ratio of the remnant liver tissue was significantly higher in the treated group. Oxidative stress imposed by PFE during the early phase of the liver regeneration enhances the Nrf2 mediated adaptive stress response leading to upregulation of GCSc gene expression and increased glutathione biosynthesis with implications of these events in enhancement of the hepatocyte`s proliferation capacity.
