Nonspecific immunoprophylactic effect of Parapoxvirus ovis preparation in a model of Aujeszky's disease virus (ADV) infection in swine

The virulence of ADV depends heavily on its ability to productively infect porcine cells of the immunohematopoetic system that may impair their immune response and might also play a role in the virus-induced immunosuppression of infected pigs. With these facts in mind we have studied systemic and local immune responses in pigs nonspecifically immunized (at Day - 4, - 2, and 0) with 2 ml of inactivated Parapoxvirus ovis, recommended by manufacturer (Bayer, Leverkusen, Germany) as a new formulation of an immune response modifier (IRM) termed Baypamun-N (BPMN), and inoculated (at Day 0) with 10^5.8 TCID_50/ml of NIA-3 strain of ADV. The obtained results are encouraging in regard to the immunoprophylactic and immunostimulatory effects of BPMN. In the infected pigs the preparation reduced/prevented clinical signs of the disease, such as body temperature and weight loss, respectively, and decreased virus shedding between postinoculation days (PID) 7 and 11. BPMN increased the in vitro responsiveness of peripheral blood lymphocytes to mitogens (PHA and PWM) between PID 7 and 11, but did not affect specific antibody production. Hence, an increase in the number of CD4a^+ helper and CD8a^+ cytolytic T cells, but not in the number of CD1^+ B cells was observed. Bronchal lymph node lymphocytes from BPMN-treated pigs exhibited moderately to slightly increased specific cell cytolysis (for 32.3% at PID 4, 8.3% at PID 11, and 24.3% at PID 14) against autologous target PHA-lymphoblasts. Thus, the use of BPMN, rather as an adjuvant (along with vaccine) than as an IRM, would be more appropriate approach for enhancing both nonspecific and specific immune responses of pigs against ADV infection.
