Comparison of different methods for enumeration of heterotrophic bacteria in aquaculture
The potential impacts of aquaculture are wide, ranging from aesthetic aspects to direct pollution problems. The degree of impact from the cage aquaculture is dependent on the species, culture method and feeding type, as well as on the nature of the receiving environment. Heterotrophic bacteria play an important role in the aquaculture environment. They are an important indicator of eutrophication. The literature presents different methods and different media for the isolation and enumeration of heterotrophic bacteria. It is a known fact that a small number (about 1%) of bacteria can be cultured on artificial media. The aim of our study was to compare two methods for enumeration of heterotrophic bacteria, spread plate method using two media for isolation and supstrat method. The study was conducted on three sea bass farms in the the South, Middle and North Adriatic Sea during two years. Water samples were collected at three depths at surface (0.5 m), subsurface (4 m) and 0.5 m from the bottom at fish farm and at control site (cca. 500 m) during the spring and autumn period. Samples were taken using 5 L Niskin sampler in sterilized plastic 0.5 L bottles. All bacterial analyses were performed in duplicate. Water samples prior to the analysis were serially diluted using sterile Phosphate Buffered Saline (PBS) (Merck). Serially diluted samples were inoculated by spread plate method on the Marine agar (Difco-BD) and Tryptic soy agar (DB-BBL) with added NaCl, as well as using substrate method, SimPlate® (IDEXX). After incubation at 35 °C during 24 h and at 22 °C during three to five days, isolated colonies were enumerated. Statistical analyses were done using SigmaStat statistical package and Statistica for Windows version 8.0. According to obtained results and performed statistical analyses number of heterotrophic bacteria at 22 °C was significantly higher determined using SimPlate test, whereas at 35 °C differences was not statisticaly significant for SimPlate test.
