Regulation of cytolytic activity of decidual lymphocytes: role of cytokines IL-2 and IL-15
During pregnancy decidua is the maternal tissue in the closest contact with the feto-placental unit tissue. Heterogeneous cell populations present in the first trimester pregnancy decidua (lymphocytes, macrophages, stromal cells and gland epithelial cells) have the capacity to secrete broad panel of cytokines. The aim was to investigate the effects of IL-2 and IL-15 which are known as modulators of cytolytic cells. Influence on cytolytic potential of decidual large granular lymphocytes (dLGL) and peripheral blood lymphocytes (PBL) of the same women was investigated. The prevalent phenotype of dLGL is CD3-CD16-CD56+, and in their cytoplasmic granula cytolytic molecule perforin (P) is abundantly present (P bright). In experiments in vitro we determined the P content in IL-2 or IL-15 stimulated dLGL and PBL, and detected the cytolytic activity of these cells against NK sensitive K562 cell line in 2hrs cytotoxicity PKH-26 (red) test. Marked differences in effects of two cytokines were observed. P expression in dLGL can be upregulated by IL-15, but not by IL-2 stimulation. Both cytokines however strongly upregulated P expression in cultured PBL. PBL stimulated by IL-15 are more cytotoxic than unstimulated cells. Contrary, cytolytic activity of IL-15 stimulated dLGL was unchanged.
