Decidual macrophages regulate cytolytic potential of decidual natural killer cells
There are very few data about the basic mechanisms regulating perforin (P) expression and cytotoxicity at maternal-fetal interface eventhough the significance of local events are emphasized. P-containing CD3-CD16-CD56bright+ decidual lymphocytes (DL) are surrounded by various cell types building the decidua including macrophages. Nonadherent DL and decidual adherent cells (DAC) were isolated from the suspension of decidual cells obtained by trypsin digestion of the first trimester normal human pregnancy decidua. Phenotype of DAC was analysed by flow cytometry and immunocytochemistry. Decidual macrophages were HLA-DR+ (activated). HLA-DR depleted and HLA-DR non-depleted cells were used as possible modulators of P expression and cytotoxicity of DL against K562 cells. Cytotoxicity was measured by PKH-26 (red) 2hrs cytotoxicity test and the percentage of killed cells was measured by flow cytometry. HLA-DR positive macrophages, present 10-20% in DAC, keep the level of perforin in 18hrs cultured DL and modulate their cytotoxicity against K562 cells. In conclusion macrophages in close contact with DL or with cytokine production in a paracrine manner, could influence perforin expression and cytolytic activity of DL.
