The influence of progesterone in regulation of perforin expression and cytolytic activity of decidual lymphocytes

Human first trimester pregnancy decidua, characterised by high local concentration of progesterone, is infiltrated by decidual leukocytes, among which non-classical NK cells are predominant cell population (85-90%). These cells are CD45+CD56bright+ phenotype and contain high level of perforin (P) in their granule. Our previous results showed that progesterone in high concentration directly downregulate the number of P+ cells as well as the content of P per cell. Progesterone indirectly in a concentration dependent manner and its mediator Progesterone Induced Blocking Factor (PIBF) block upregulation of P expression in decidual lymphocytes (DL) cultured with decidual macrophages. The aim of this study was to investigate the role of progesterone in the regulation of cytotoxicity of DL. Distribution of PIBF+ cells was shown by immunocytochemictry of paraffin embedded decidual tissue of first trimester normal human pregnancy. The suspension of freshly isolated decidual cells was incubated overnight with or without progesterone and cytolytic activity of DL was measured against K562 cell line in 2 hr cytotoxicity PKH26 (red) test. Our results point that strong progesterone/PIBF influence at materno - fetal interface seems to be crucial for regulation of P expression and suppression of cytotoxic activity in decidua.
