Relevance of paraoxonase-1, platelet- activating factor acetylhydrolase and serum amyloid A in bovine mastitis

Bovine mastitis is the most economically important production disease in dairy cows. The pathogenesis of mastitis involves an inflammatory reaction resulting from production of cytokines at the site of infection. Serum amyloid A (SAA) is one of the major APP in ruminants whose concentration rises dramatically during acute phase response (APR). Native HDL contains paraoxonase-1 (PON1) and platelet-activating factor acetylhydrolase (PAF-AH), both antioxidative/anti-inflammatory enzymes being responsible for anti-inflammatory/anti-oxidative properties of HDL. In order to evaluate systemic inflammatory and oxidative stress response, we investigated serum amiloid A (SAA) concentration, PON1 and PAF-AH activities in serum of cows with subclinical and clinical mastitis. Our results showed that the inflammation interfered with lipid metabolism affecting concentration of total cholesterol, HDL-C and triglycerides (TG) in cows with clinical mastitis. The results demonstrate that SAA concentration in the serum is a reliable marker for diagnosis of clinical mastitis showing high response in cows with clinical form of disease but could not distinguish subclinical from clinical mastitis. On the other hand, lower PON1 activity in subclinical mastitis with moderate response demonstrates PON1 as a possible marker for diagnosis of subclinical form of disease.
