TH1 cytokines in the regulation of cytotoxicity at the interface
Cytolytic cells, both cytotoxic T lymphocytes (CTL) and Natural killer (NK) cells are endowed with the potent cytolytic machinery. Decidual NK cells (dNK) are predominant lymphocyte population at the maternal-fetal interface in humans. It has been suggested that dNK besides functions in the defense against virally infected or otherwise parasitezed cells and transformed cells may contribute to the control of cytotrophoblast invasion of the placental bed during early pregnancy. For these functions dNK cells are abundantly endowed with the perforin (P bright) which is the molecule mediating cytotoxicity at the molecular level. It is a general opinion that the pregnancy is a Th2 phenomenon and the prevalence of Th2 type cytokines suppress cytotoxicity. However the role of Th1 type cytokines could be very important in the protection of feto-placental unit against both maternally acquired and local infections at the interface. It could be supposed that in these circumstances response at the interface is biased towards Th1 type cytokines. We have investigated the role of Th1 type cytokines (IL-2, IL-12, IL-15 and IL-18) in the control of the level of P expression and NK cytotoxicity of decidual lymphocytes. Cytotoxic potential of DL is highly increased by high doses of IL-2 (1000 IU/ml) and IL-12, but surprisingly suppressed by IL-15. This suppression is released in the combination of other Th1 type cytokines. These results are clearly showing that at the interface cytolytic pathway is ready for functions but its activation is controlled. However, in pathological conditions (Th1 type cytokines prevalence) cytolytic pathway can be triggered.
