Progesterone and progesterone induced blocking factor (PIBF) regulate perforin expression and cytolytic activity of decidual lymphocytes

Non-classical NK cells are predominant cell population in first trimester normal human pregnancy decidua. These cells are CD45+CD56 bright+ phenotype and contain high level of perforin (P) in their granules. Besides rich infiltration of leucocytes, decidua is characterised by high concentration of progesterone (3g/g of tissue). Our previous results showed that progesterone in high concentration directly downregulates the number of P+ cells as well as the content of P per decidual lymphocytes (DL). Progesterone indirectly in a concentration dependent manner, and its mediator Progesterone Iniduced Blocking Factor (PIBF), block upregulation of P expression in decidual lymphocytes cocultured with decidual macrophages. The aim of this study was to investigate the distribution of PIBF+ cells and the possible role of progesterone in regulation of cytotoxicity in decidua. Distribution of PIBF+ cells was shown by immunohistochemistry of paraffin embedded decidual tissue of first trimester normal human pregnancy. The suspension of freshly isolated decidual cells was incubated overnight with or without progesterone or PIBF and cytolytic activity of DL was measured against K-562 cell line in 2 hour cytotoxicity PKH-26 (red) test. Our results point that strong progesterone/PIBF influence at materno-fetal interface could be crucial for regulation of P expression and suppression of cytotoxic activity in first trimester normal human pregnancy decidua.
