Is the Simple Calibration Accurate Enough for Spectrophotometric Screening of Total Carotenoids in Corn Flours?
Different calibration approaches when applied to spectrophotometric analysis of total carotenoids (TC) in yellow corn products were compared. These were calibration with beta carotene (BC) or TC standard in hexane or in a real blank, and additions of BC standard to the samples. The usefulness of the aforementioned approaches for reliable analysis of TC in yellow corn flours and grits was fully evidenced. Data obtained pointed to comparable accuracy, sensitivity, and imprecision of these approaches. All this makes them equivalent and mutually interchangeable. From the point of simplicity and cost-effectiveness, the calibration in hexane is suggested. Laborious and demanding approaches such as standard additions and calibration in a real blank are redundant and may be replaced by a simple calibration in the solvent. Therefore, the answer to the title question is yes ; the analyst is allowed to use simple calibration in hexane with BC or TC when screening large number of samples in breeding experiments or other routine trials.
