The electric impulses and human interferon-alpha affects growth properties of cellular "microtumors" in vitro
Exposure of cells to an electric/electromagnetic field produces changes in the synthesis and secretion of different molecules and induces the intracellular metabolic changes. The influence of electric impulses and human interferon-alpha on the growth properties of "microtumors" in vitro of WIREF and HeLa cells was studied. Monolayers in Eagle's medium+10% of FCS were cultivated. To get the "microtumors", cells were seeded into the "bacteriological grade" Petri dishes. After 3-4 days at 37^oC in CO_2 atmosphere, the monolayer cells begin to form the "microtumors" consisting of 30-100 cells. They were sedimented at 500 RPM/10 min. and re-suspended in fresh medium+FCS in a "conc." of cca. 1000 "microtumors"/ml. and exposed to: (1) Direct square-wave impulses of 10 micros duration with 10s break, with a field force of 100 V/cm ; (2) Capacitive impulses at 290 V 1-3 nF of 10micros duration with 10s break ; (3) 10/1000 l.U. /ml of HuIFN-alpha. "Microtumors" were seeded into the tissue-culture grade dishes, and in 3-5 days the atachment and spredding on the surface was followed. AF (alkaline phosphatase) and the NO in the medium were analysed. The data shows: (1) Different effect of inductive and capacitive electric impulses on WiREF and HeLa cells "microtumors" in sense of no. of impulses. (2) 6-9 impulses are needed to inhibit the attachment and growth of microtumors. (3) HuIFN-alpha did not influenced the growth and attachment when added alone. It was effective in combination with 6-9 of inducitve or capative impulses. (4) The level of NO (Nitric oxide) in the medium was reduced after 6-9 impulses only.
