Validation of thin-layer chromatography quantitation determination with CCD camera and slit-scanning densitometer
Video densitometric and classical slit-scanning determination of pesticides on TLC plates were validated for linearity, precision and detection limit. A comparison of results obtained showed that slit-scanning densitometry is definitely more sensitive and more precise than video densitometry. However, according to the validation requirements, RSD of video densitometry is in the acceptable range (3.5-5.3 %). The linearity is very good and almost identical for both techniques. The main advantage of video technology is the measuring velocity (a few seconds for the measurement cycle compared to 20 minutes with scanning densitometry) and excellent archiving facility, so that the image and chromatographic data can be stored together, edited and used for many tasks.
