Peroxidase activity in selenium and copper treated liver

Selenium and copper as essential elements control the activity of certain enzymes that are part of cellular defense mechanism. Ingested copper accumulate particularly in the liver in metal inducible protein metallothionein-ceruloplasmin which functions as a ferroxidase. Selenium is cofactor in enzyme glutathione peroxidase which is included in the cell antioxidant system. The aim of the present work was to investigate effect of prolonged contamination with low concentration of selenium and copper on expression of peroxidatic enzyme activity. Research was conducted on carp (Cyprinus carpio L.) treated with low concentrations of Se (0,1 mg/L) andCu (0,1 mg/L) during sixteen days. The liver samples were taken for analysis every day during first four days, and after that period, in intervals of two or three days. The peroxidase activity was determined spectrophotometrically by measuring the values of absorbency at 470 nm. Results showed that the activity of peroxidase were rapidly increased during first day of treatment. Immediately after that period activity was decreased. On the fourth day of treatment values were approximately equal or even slightly lower than in the begining of treatment. With prolonged treatment values of peroxidases were increased again but with oscilations which did not permit significant abberation from values of control animals. However, it was evident that in both cases of contaminated animals peroxidase activity showed time congruity with raises and falls, which indicated that the cellular response was uniformed to a presence of metals.
