NMDA blockers and the brain free fatty acid level in rats with status epilepticus induced by lithium and pilocarpine administration
Seizures trigger a variety of biochemical processes including an influx of extsacellular Ca2+ through voltage-dependent and glutamate-operated channels, activation of membrane phospholipases, liberation of free fatty acids (FFA), diacylglycerols, eicosanoids, lipid peroxides and free radicals. These lipid metabolites along with abnormal ion homeostasis may be involved in cell injury and cell death. The aim of this study was to determine the influence of the NMDA channel blockers, ifenprodil and MK-801, on the brain FFA level in rats with generalised convulsive status epilepticus induced by lithium and pilocarpine administration. Experiments were carried out on Hannover-Wistar rats. Control group of animals was intact and had no past experimental history. All other rats received lithium chloride (3 mmol/kg i.p.) and 20 hours later pilocarpine (30 mg/kg i.p.). Vehicle solution (ethanol : propylene glycol 400 (1:1 V/V) or ifenprodil (1, 3 or 10 mg/kg) or MK-801 (0.3 mg/kg) had been injected i.p. 30 min after administration of pilocarpine. All rats were decapitated 30 min after the administration of vehicle solution, ifenprodil and MK-801. The brains were quickly removed and frozen in liquid nitrogen. The FFAs were quantified using gas chromatography method. The results demonstrate that lithium plus pilocarpine - induced status epilepticus causes the brain FFA accumulation. Tested dose of MK-801 did not influence the increase of the brain FFA level, while ifenprodil in tested doses prevented the accumulation of the brain free palmic, stearic and oleic acid in rats with status epilepticus.
