Calcareous sponges (Porifera, Calcarea): genetic analysis and evolution

Calcarean phylogeny plays an important role in understanding of early metazoan evolution, due to their closer relationship to Eumetazoa than to Demospongiae and Hexactinellida. The Calcarea has a reputation of being obscure and taxonomically difficult because these animals offer very few morphological characters for reliable phylogenetic reconstruction. Many different species were lumped into synonymy, as their diagnostic morphological differences were reinterpreted as a result of intraspecific variability. Recent morphological and molecular analyses have demonstrated that enormous morphological variability of sponges is not the rule and that highly variable and widely distributed sponge species were not as common as previously thought. Together with detailed histology, molecular data appear to be essential to provide substantial evidence for phylogenetic hypotheses based only on molecular methods. This study presents an effort to inventory the markedly underestimated calcarean sponge diversity in the Adriatic Sea, thus introducing five species new to science. We combined traditional morphological approach (spicule size, shape and distribution) and molecular (using partial 18S, 28S ribosomal DNA sequences, and ITS1-5.8S-ITS2) analysis using individuals of class Calcinea and Calcaronea. Our results extended the currently available dataset of Calcarean species, showing high biodiversity in the Adriatic Sea and showed that the taxonomy of Calcarea is in desperate need of a thorough revision.
