Temperature changes measurement during electroporation
We have developed a thermocouple temperature measurement system for precise and fast measurements of the temperature, which allows measuring temperature changes in the tissue during electroporation. The temperature is measured using a thin (80 µm in diameter) T-type thermocouple wire. Signal conditioning is performed by the integrated circuit AD595 that already implements cold junction compensation and amplification of the thermocouple signal. The amplified signal can be sampled using an oscilloscope. To validate the functioning of the developed system, measurements of temperature increase in animal liver during electroporation were performed. Moreover, we performed additional simultaneous measurements using a commercially available optical temperature measurement system which was also connected to the oscilloscope to perform a direct comparison of both measured temperatures. The thermocouple thermometer we developed is fast device due to small thermocouple probe. The device can measure temperature changes during the electric pulses which allow us to validate the mathematical model of the tissue heating during the electroporation.
