Lysosomal membrane destabilisation as marker of cellular stress in freshwater invertebrates

Lysosomes concentrate a wide range of contaminants from within the environment, including lipophilic xenobiotics and metals, what may cause changes in the integrity of their membranes. Retention of the cationic probe neutral red within the lysosomal compartment over time was used as a measure of damage to the lysosomal membrane. The acid environment of lysosomes is maintained by the membrane Mg2+-ATPase dependent H` ion proton pump. The neutral red retention assay (NRR) reflects the efflux of the lysosomal contents into the cytosol following damage to the membrane and, possibly, impairment of the I-f` ion pump. The objective of this research was to investigate possible use of haemocyte lysosomes of freshwater molluscs as cellular marker of environmental pollution. In our research we used haemocytes of freshwater mussel Dreissena polymorpha Pall. and freshwater snails Lymnaea stagnalis L. and Planorbarius corneus L. Cadmium, copper and pentachlorophenol were used as reference compounds. Also, exposition of mussels D. polymorpha was conducted in River Sava, downstream from Zagreb. Our findings confirm that NRR can be used as rapid, easy and cost-effective marker of freshwater pollution.
