The assessment of DNA damage in poultry spermatozoa after exposure to low doses of ionising radiation

The assessment of DNA damage in poultry spermatozoa after exposure to low doses of ionising radiation using comet assay (alkaline and neutral) was not previously reported. The aim of the study was to investigate the effects of different doses of gamma-radiation on tail length (TL), tail intensity (TI) and tail moment (TM) of cocks’ spermatozoa using comet assay. Semen samples of 5 cocks of heavy line (45 weeks old) were collected by dorso-abdominal massage method. All samples were diluted and irradiated in vitro with doses of 0.3, 0.5, 1 and 2 Gy. One-way ANOVA was used to compare differences in DNA damage. Results showed high sensitivity of the method even at low dose exposure from 0.3 Gy, with no dose related relationship, but with increase of apoptotic cells. Damage distribution was spread more after 0.5 Gy, 1 Gy and 2 Gy dose. TI and TM were lower than in fresh semen cock samples (other researchers), while TL was little bit higher. Lower damage for higher doses 0.3 Gy was strange result, and since semen samples do not have DNA repair mechanisms, further larger studies are needed to answer that question.
