Elucidation of metal ion-binding sites in proteins
Metal ions are constituents of about 40% of proteins1 where they can act as structure promoters or can take part in enzymatic reactions. Removal or replacement of one metal by another is often accompanied either by loss, or reduction, or even alternation of the enzyme catalytic potency2. Apparently knowledge of the type and number of electron donors is important for modeling an enzyme catalyzed chemical reaction as well as to clarify how specific certain metal binding sites are. We have combined the web-based platform for analysis of metal-binding sites in proteins3 and quantum mechanical – DFT calculations to reveal details of the metal binding sites in several enzymes. Herein I’ll summarize results of our theoretical studies on two dioxygenase4 and one dipeptidase.
