The role of IL-15 on the maintaining of perforin protein in decidual and peripheral blood lymphocytes

The decidua of pregnancy is the maternal tissue in the closest contact with feto-placental unit tissues. Cell populations in the decidua are heterogeneous containing gland epithelial cells, fibroblasts, macrophages, lymphocytes and other types of stromal cells. All these cells are very close one to each other and many are capable of secreting cytokines which can influence other cells in a paracrine manner. It is a general opinion that an immune-endocrine network is operating at the maternal-fetal interface. In experiments in vitro we have shown that decidual adherent cells (DAC) are involved in regulation of perforin (P) protein expression in decidual non adherent cells (DNAC) by preventing downregulation of P expression in 18h culture. DAC were more powerful in this function than peripheral blood adherent cells PBAC (20% of DAC added had the effect comparable to 50% of PBAC). The aim of this investigation was to analyze whether this effect is mediated by cytokines IL-12 and IL-15. The results showed that neutralization of IL-12 effects on DNAC and PBNAC were without any influence on the level of P protein expression. However, by addition of anti-IL15 antibody the effects of adherent cells on both DNAC and PBNAC perforin expression can be blocked, at least partially. Anti IL-15 antibodies were efficient also in blocking upregulation of perforin expression with supernatant obtained by cultivation of DAC. In our opinion direct and indirect (cytokine and hormone mediated) interactions between DAC and DNAC is responsible for high level of P expression in the decidual lymphocytes. In the experiments described the role of IL-15 is emphasized. These investigations are financially supported by the Ministry of Science and Technology of the Republic of Croatia (Grant NO. 062001).
