Perforin mRNA expression in human first trimester pregnancy decidual and peripheral blood lymphocytes
Background. There is an increasing interest recently in the role played by lymphocyte mediated cytotoxicity at the materno-fetal interface. Abundance of perforin (P) expression in the granules of lymphocytes infiltrating the decidua of pregnancy is pointing to the role of secretory cytolytic pathway. However, the precise role of that strong cytolytic machinery is still far from being understood. The aim of the experiments presented herewith was to study mechanisms of P gene expression at the mRNA level in normal first trimester pregnancy decidual (DL) and peripheral blood (PBL) lymphocytes. The reverse transcription polymerase chain reaction (RT-PCR) was used to study gene expression.Methods and results. Decidual lymphocytes were obtained by trypsin digestion from elective first trimester termination of pregnancy, and PBL'S from same individuals were used. Suspensions of PBL and DL, obtained by Ficoll-Hipaque gradient centrifugation, was incubated overnight in tissue culture Petri dishes to obtain non-adherent (NAC) and adherent cell (AC) population. Total cytoplasmic RNA was extracted and reverse transcripted into cDNA and than amplified with PCR. Products were quantified in normalization to GAPDH gene. Decidual adherent cells (DAC) maintain mRNA for perforin both for DL and PBL in all times of incubations (0h, 1h, 3h, 6h, 18h). When adherent cells were removed, mRNA for perforin was absent in 0h and 1h, peak was in 6h and slight decline in 18h for DL. In PBL mRNA for perforin was present in 0h and 1h, have a peak in 6h and slight decline in 18h. Stimulation with PMA showed no detectable mRNA for perforin in all times of incubation both for DL and PBL. Conclusion. In spite of being preliminary results are showing that P gene expression, studied at the mRNA level, is different in DL and PBL, and that adherent cells participate in the control of P gene expression.
