The role of RSC complex in chromatin structure remodeling at the yeast PHO5 promoter

PHO5 promoter of Saccharomyces cerevisiae is a classical model for studying mechanisms of chromatin structure remodeling as a prerequisite for promoter activation. There is a redundant network of chromatin remodeling pathways involved at the PHO5 promoter, but no essential chromatin remodeler has been identified. RSC is the only chromatin-remodeling complex essential for cell growth, whose role at the PHO5 promoter remained unresolved. In this study a viable deletion mutants of specific structural subunits of two distinct RSC complex isoforms, Rsc1 and Rsc2, as well as conditional temperature sensitive mutant of the RSC ATPase subunit, sth1td, were used to elucidate possible role of RSC in the PHO5 promoter activation. Inactivation of Rsc2 subunit or depletion of catalytic subunit Sth1td, strongly delayed but not prevented promoter chromatin structure remodeling. Combined inactivation of both Rsc2 and Sth1 prevented chromatin opening and consequently transcriptional activation. The same was observed with the PHO5 promoter variant activated through GAL signaling pathway. The results pointed out an essential role of the RSC complex in a chromatin remodeling process at the PHO5 promoter.
