Low level of mozaicism in couples with recurrent molar pregnancies.
Most of the complete hydatidiform moles were caused by fertilisation of an empty egg by a duplicated haploid sperm, but rare exceptions may exist. Molar degeneration is caused by paternal uniparental diploidy, it means that all chromosomes of molar tissue have paternal origin. Recurrence suggests a strong genetic predisposition. In families with extensive intermarriage and recurrent molar pregnancies, unusual MHC class I and II antigens were found, but the reason for the repeated reproductive failure or striking difference in frequency of disease in different population is not known yet. Three women suffered from recurrent hydatiform molar pregnancies and their spouses, were analyzed cytogeneticaly. Low level of mosaicism 45,X was found in female and marker chromosome in male partner. We also analyzed skin fibroblast from one male partner. The low frequency of mosaicism reported here would not be detectable by conventional chromosomal analyzes of 10 mitosis per person. The implication of mosaic karyotypes in causation of molar pregnancy will be discussed.
