Photochemical synthesis of medium and large aza-heterocyclic compounds with potential biological activity
Various aza-heterocyclic compounds show biological activity. For example, pyrrolo[1, 4]benzodiazepines show highly selective binding to DNA.1 Likewise, benzazepine skeleton is a part of numerous biologically active compounds.2 Furthermore, adamantane is a very important pharmacophore that has been found in different classes of biologically active compounds.3 Therefore, it is necessary to develop new synthetic methods for the preparation of new functionalized adamantyl-heterocyclic derivatives. With the ongoing interest in our group to apply photochemical approach to the synthesis of adamantyl-heterocyclic derivatives with biological applications, 4 we have functionalized adamantanes by the phthalimide group, versatile chromophore that undergoes various photochemical reactions. Herein we describe synthesis of novel (N-phthalimido)adamantane carboxylic acid 1 and its efficient one-pot transformation to complex heterocyclic compound, adamantyl benzazepine 2, with anticipated biological activity, that is yet to be tested.
