CD34+ and CFU-GM content during peripheral blood CD34+ selection for autologous transplantation
The aim of the study was to analyze the relationship between the CD34+ and CFU-GM content in leucocyte products during CD34+ selection (Ceprate, CellPro) in patients undergoing autologous transplantation. A total of 37 patients were analyzed, including 12 pts. With non-Hodgkin lymphoma, 11 with breast carcinoma, 10 with multiple myeloma, 3 with Mb. Hodgkin and 1 pt. with ovarian cancer. Mobilization protocol consisted of cyclophosphamide, BEAM or VIP-protocol, all in combination with G-CSF. Leukaphereses contained a median of 6.95x10E6 CD34+/kg b.w. (2-7-40.6) and 20.1x10E4 CFU-GM/kg b.w. (1.1-117.8) (r=0.412, p<0.05). The median CD34+ purity in selected product was 83% (28-96), whereas the calculated yields of CD34+ cells and CFU-GM with regard to apheresis product were 36.2% (2-7-82.2) and 15.1% (3.4-49.6), respectively. Although CD34 and CFU-GM were significantly correlated, no correlation was Found between the extent of CD34+ and CFU-GM decrease (r=0.057), indicating a different rate of specific cell loss during the CD34+ selection.
