Fluidity of the myelin sheath is changed in the peripheral nerves of diabetic rats

The peripheral neuropathy associated with diabetes is one of the most common neuropathies. The characteristical histopathological findings are axonal degeneration, secondary demyelination and athrophy. The underlaying pathological mechanisms are still unclear, but among possible factors involved are protein glycation, altered lipid metabolism, decreased inositol content, altered Schwann cell function, local ischemia, and lack of neurothrophic factors. In the present study we examine structural integrity of myelin sheath in the peripheral nerves from short-term streptozotocin (STZ)-diabetic rats, using electron spin resonance (ESR) as a tool in determining the dynamic state and structure of myelin lipid phase. Experiments were performed on sciatic and sural nerves of STZ-treated rats and age-matched controls. Prepared nerve samples were spin labeled by lipophilic spin probe of MeFASL(10, 3) and respective ESR spectra recorded. The spectrum analisys was performed using a numerical simulation model with the set of fitting parameters that in the same time relate the ESR line shape and structure and dynamics of the probed environment. The analisys considered three typical spectral components weighted and summed in the composite spectrum. The analisys of the experimnetal spectra showed that although the values of the fitting parameters of the spectral components did not change significantlly, the weighting factor for the spectral component II was elevated significantlly for te diabetic samples. This indicated the increase in the overall fluidity of the myelin shetah lipid phase in peripheral nerves of the STZ diabetic rats. Possible mechanisms of such an effect are discussed.
