The influence of the C(3)-substituent on chemical transformations of 1, 4-benzodiazepin-2-ones upon Cu(II)-coordination

1, 4-Benzodiazepin-2-one derivatives are well known CNS active compounds. Well documented properties of nitrogen ligands to form metal complexes possessing biological activity even higher than the free ligands, prompted us to prepare novel copper complexes of 3-hydroxymethyl- (1), 3-acetoxymethyl- (2) and 3-methyl- (3) derivatives of 7-bromo-1, 3-dihydro-1-methyl-2H-1, 4-benzodiazepin-2-one. The synthesis, some spectroscopic properties and X-ray structure analysis of free ligands as well as the copper(II)dichloride complexes are presented. During the crystal growth of complexes, however, unexpected chemical transformations occurred. Mechanisms of these chemical transformations are proposed. The complexes were prepared by reaction of copper(II)chloride dihydrate and benzodiazepine ligands in 1:1 metal to ligand ratios in different solvents: ligand 1 in absolute ethanol whereas ligands 2 and 3 in a 1:1 mixture of dichloromethane and ethanol. In the first case (ligand 1) two kinds of crystalline complexes were obtained by slow evaporation of ethanol at the room temperature: complex B (green) and complex C (black) which are, as shown by the X-ray structure analysis, products of oxygenation (B) and subsequent oxydative ring contraction (C) of 1. In the case of 2, upon copper coordination a green [CuCl2(2)] was afforded (complex D). Only after heating, dark brown crystals are formed, which are shown to be a quinazoline derivative copper complex (E), a product of the oxydative ring contraction similar as encountered by complexation of 1. However, chemical transformation of the ligand was not detected for complexation of copper to 3, only a green [CuCl2(3)] complex (F) was formed.
